BBI Solutions’ Alkaline Phosphatase (ALP) is extracted from bovine intestine then purified using multiple precipitation, fractionation, chromatography and filtration steps.

ALP is commonly used as a label in immunoassays such as ELISA, and in blotting and histochemistry. Once conjugated to antibodies, antigens, or streptavidin, its low backgrounds and linear reaction rate enables increased sensitivity over extended incubation times. It can be used with a variety of substrates producing precipitated or soluble chromogens, or with chemiluminescent and fluorogenic substrates for enhanced sensitivity.

Our high activity conjugation grade products are supplied as glycerol solutions.

We recommend you store the material between 2 - 8°C. This product is shipped at ambient temperature.

BBI grade ALPI12G, which has the highest specific activity currently available.

BBI’s Glucose Oxidase is produced from large-scale fermentation of the fungus Aspergillus niger. The crude fermentation liquor is purified using multiple precipitation, fractionation, chromatography, and filtration steps.

Glucose Oxidase is used in liquid and powder glucose laboratory reagents, urine test strips, colorimetric blood glucose strips and for blood glucose monitoring in biosensors.

Our final products are lyophilised powders.

BBI’s urease is obtained from specially selected and cultivated jack beans. The enzyme is extracted from the beans then purified using multiple precipitation, fractionation, chromatography and filtration steps.

Urease is used in liquid and powder urea laboratory reagents, and biosensors. It can be used in the determination of urea in biological fluids (couple with GLDH, in the ultraviolet method). Urease can also be used as an enzymes label in ELISA.

Enzymes for Immunoassay 

At BBI Solutions, we aim to provide you with all of the biological raw materials that you need to develop and manufacture ELISA assays and other immunoassay based diagnostic tests. Most immunoassays involve linking a detectable label such as an enzyme to an antibody or antigen, which produces an observable colour change in the presence of certain reagents. 

The properties of enzymes as reaction catalysts, i.e. speed and specificity, make them excellent tools for assay end detection when combined with suitable substrates. Use of enzyme labels allow assay optimisation through sensitivity, convenience and speed of reactivity, and have facilitated the development of several widely used test formats for the same analyte. 

Horseradish Peroxidase (HRP) and Alkaline Phosphatase (ALP) are the most commonly used as enzyme labels to provide signal amplification in the majority of the immunoassays made and used today. 

Key properties for Enzymes used in Immunoassay 

High activity and purity 

A high enzyme activity ensures that the sensitivity of the assay is limited by the characteristics of the antigen to antibody binding rather than the ability of the enzyme to determine immune reactivity.  

Specific activity 

Enzymes used for detection should have a substrate specificity different to that of any enzymes present in test samples, thereby not producing a detectable signal with any components of those samples e.g. serum.

Easily conjugated 

For ease of manufacture, enzymes should have the potential to be linked to a range of proteins and other analytes by relatively simple and established methods that do not interfere with the enzyme reactivity. 

Stable 

When storing, the purified enzyme and enzyme conjugates should be stable in a wide range of formulations; room temperature stability being a distinct advantage for many applications. 

Readily available & inexpensive 

Commercial preparations must be consistently available at high purity and specific activity, and at prices not contributing significantly to the final cost of the test. 

Key Enzyme Labels 

Horseradish Peroxidase 

Horseradish peroxidase (HRP) is one of the most commonly used enzyme labels in immunoassays. 

The purity of the HRP preparation can be indicated by determination of the Reinheitszahl ratio (Rz) and preparations suitable for use in labelling should have an Rz of 3.0 or greater. 

There are many HRP Isozymes, and it is generally accepted that Isozyme C has the highest specific activity, and this can account for up to 40% of the peroxidase content of horseradish. However, several of the isozymes with “neutral” isoelectric points have specific activities perfectly adequate for use as labels in high sensitivity immunoassays so it is not necessary to use the highest purity, and therefore most expensive, Isozyme C preparation. 

In some assays, even when using highly purified HRP, it is possible to get background signal. In such cases an improvement to the “signal/noise” ratio can be obtained by inclusion of purified but inactive Apo HRP.

Alkaline Phosphatase 

ALP is the other enzyme of choice for the majority of antibody conjugations. It is often used as an alternative to HRP when a more linear reaction rate is required over a wide range of values. The source of ALP with the highest specific activity, and therefore of most use in assays, is calf intestine. 

BBI’s ALPI12G is our highest specific activity product and bestselling line. Its performance has been proven in assays manufactured by some of the largest immunoassay and IVD companies.
